Understand the script and what they want - although this is the same no matter the audition, if you are on your own actually having intention and committing to the ideas you come up with is a tough lesson to learn.
Personally (and I think that it's only me.), I get everything I need to do in one take. My Spouse is my camera/sound/director (lucky for me, a job she also did in TV so I know I can trust that. Perhaps another tip?), and I don't want to keep her for too long, so set it all up, rehearse, ready yourself, call the Director in the room, let them know what you are 'trying' to do and do 1 take. If they like it, you are done. If not, they'll tell you what changes to make. Do it again with all of this onboard and that's it.
If you commit to no more than 2 takes your concentration should see you to a good take.
