I would have to lean towards films where the title is said in the film at some point, and how it moves me when it's said. So from that viewpoint, for me the Single Greatest Film Title of ALLLLLLL Time would have to be Once Were Warriors. By the time Beth (played by the great Rena Owen) says those words in that film....oh man...I died a little bit. That is such a great film and she is such a great actress. It's painful that she didn't win Best Actress in her country for that role.
As Good As It Gets; Cool Hand Luke; Chinatown; Silence of the Lambs and Kajillionaire are in the same vein and are all probably tied for second.

The Hitman's Bodyguard. I love the action mixed with humour. How disgusted the bodyguard (played by Ryan Reynolds) was in that scene where the hitman smashes the window of the car... Ahhh, the reaction of his bodyguard was fabulously timed. I can't wait for 'The Hitman's Wife's Bodyguard', ' Free Guy', and 'Red Notice'. I live in Germany so I have to wait longer for the movies to come out here but the wait makes it even better.
I really love Three Identical Strangers as a title, it really gets the intrigue going. I also love the title Under The Silver Lake. Haven't watched it and it sounds like it's not a great film, but there's something really beautiful about the poetry of the words, almost Dylan Thomas-esque..

I'm just going to third that "Nope" is an incredible title.
Book-wise, though: "A Heartbreaking Work of Staggering Genius" & "The Brief Wondrous Life of Oscar Wao".
